Abstract

Phytochemical screening and synthesis of silver nanoparticles (AgNPs) have been carried out using leaf extract of Seribu Kuman (Rhinacanthus nasutus). Phytochemical screening of the leaf extract of Seribu Kuman showed the presence of secondary metabolites of alkaloids, flavonoids, triterpenoids, steroids and saponins. Silver nanoparticles were synthesized by varying the extract volumes and temperatures. The best variation is determined by looking at the highest absorbance value and the maximum wavelength that appears in the 400-480 nm region on a UV-Vis spectrophotometer. The best variation was obtained at a variation of the extract volume of 9 mL and a temperature of 50 degrees celcius with an absorbance of 0,981 and maximum wavelength of 435 nm. The synthesized silver nanoparticles in the best variation were further characterized using XRD and FTIR. XRD diffractogram data of silver nanoparticles showed the typical 2 theta angle value of silver nanoparticles according to the JCPDS database No. 04-0783 at 37,95 (111); 44,30 (200); 64,53 (220) and 77,30 (311) with a crystal size of 7,970 nm and a crystal structure is Face Centered Cubic (FCC). FTIR spectrum analysis on extract and silver nanoparticles showed the loss of absorption of C=O aldehyde to C=O absorption of carboxylate ion which indicated a reduction in silver ions. The antibacterial activity test of silver nanoparticles against Escherichia coli and Staphylococcus aureus showed antibacterial activity which was included in the moderate category with an average clear zone diameter of 10,23 mm and 8,31 mm, respectively. Telah dilakukan skrining fitokimia dan sintesis nanopartikel perak (AgNPs) menggunakan ekstrak daun Seribu Kuman (Rhinacanthus nasutus). Skrining fitokimia ekstrak daun Seribu Kuman menunjukkan adanya senyawa metabolit sekunder golongan alkaloid, flavonoid, triterpenoid, steroid dan saponin. Sintesis nanopartikel perak dilakukan dengan memvariasikan volume ekstrak dan temperatur. Variasi terbaik ditentukan dengan melihat nilai absorbansi tertinggi dan panjang gelombang maksimum yang muncul pada daerah 400-480 nm pada spektrofotometer UV-Vis. Variasi terbaik diperoleh pada variasi volume ekstrak 9 mL dan temperatur 50 derajat celcius dengan absorbansi 0,981 serta panjang gelombang maksimum 435 nm. Nanopartikel perak hasil sintesis pada variasi terbaik dikarakterisasi lebih lanjut menggunakan XRD dan FTIR. Data difraktogram XRD dari nanopartikel perak menunjukkan nilai sudut 2 theta khas nanopartikel perak sesuai database JCPDS No. 04-0783 pada 37,95 derajat (111); 44,30 derajat (200); 64,53 derajat (220) dan 77,30 derajat (311) dengan ukuran kristal 7,970 nm dan struktur kristal berupa Face Centered Cubic (FCC). Analisis spektrum FTIR pada ekstrak dan nanopartikel perak memperlihatkan hilangnya serapan C=O aldehid menjadi serapan C=O ion karboksilat yang menunjukkan terjadinya reduksi ion perak. Pengujian aktivitas antibakteri nanopartikel perak terhadap Escherichia coli dan Staphylococcus aureus menunjukkan adanya aktivitas antibakteri yang termasuk ke dalam kategori sedang dengan diameter rata-rata zona bening masing-masing sebesar 10,23 mm dan 8,31 mm.
