Abstract

Synthesis of silver nanoparticles (AgNPs) using putat leaf extract (Planchonia valida) had been carried out. AgNPs synthesis was carried out by varying the extract volume and heating temperature. The formation of AgNPs was observed by a color that change from yellow to brownish red and also confirmed by using a UV-Vis spectrophotometer which was marked by the appearance of a peak in the surface plasmon resonance (SPR) region in the range of 400-480 nm. The optimum conditions for the formation of AgNPs were seen from the absorption of the maximum wavelength of the SPR and the absorbance which indicated the number of AgNPs formed. The optimum condition AgNPs were further characterized using FTIR and XRD and tested for their antibacterial activity against Escherichia coli and Staphylococcus aureus by disc diffusion method. The optimum results of AgNPs synthesis were obtained at an extract volume of 8 mL and a heating temperature of 60oC with λmaks of 421 nm and absorbance 0.755. XRD diffractogram data shows typical 2θ angle values of silver nanoparticles according to JCPDS database No. 04-0783 at 38.32o (111), 44.00o (200), 64.59o (220), 77.32o (311) with an average crystal size of 8.7627 nm and a Face Centered Cubic (FCC) structure. The FTIR spectra of putat leaf extract and silver nanoparticles showed a loss of C=O aldehyde absorption at 1736 cm-1 to C=O absorption of carboxylate ions at 1614.42 cm-1 which indicated a reduction of silver ions. In addition, several other absorption peaks at FTIR tends to act as a capping agent. Antibacterial activity testing of silver nanoparticles showed the antibacterial ability which tended to be the same, namely the medium category against Escherichia coli and Staphylococcus aureus bacteria with diameter of clear zones of 10.29 mm and 10.03 mm, respectively. Telah dilakukan sintesis nanopartikel perak (AgNPs) menggunakan ekstrak daun putat (Planchonia valida). Sintesis AgNPs dilakukan dengan memvariasikan volume ekstrak dan temperature pemanasan. Terbentuknya AgNPs diamati dengan adanya perubahan warna dari kuning ke merah kecoklatan dan kemudian dikonfirmasi menggunakan spektrofotometer UV-Vis yang ditandai munculnya puncak pada daerah surface plasmon resonanace (SPR) yaitu kisaran 400-480 nm. Kondisi optimum terbentuknya AgNPs dilihat dari serapan panjang gelombang maksimum SPR serta absorbansi yang mengindikasikan banyaknya AgNPs yang terbentuk. AgNPs kondisi optimum kemudian dikarakterisasi lebih lanjut menggunakan FTIR dan XRD dan diuji aktivitas antibakterinya terhadap Escherichia coli dan Staphylococcus aureus dengan metode difusi cakram. Hasil optimum sintesis AgNPs diperoleh yaitu pada volume ekstrak 8 mL dan temperature pemanasan 60oC dengan λmaks 421 nm dan absorbansi 0,755. Hasil pengukuran dengan XRD memperlihatkan nilai sudut 2θ khas nanopartikel perak sesuai database JCPDS No. 04-0783 pada 38,32o (111), 44,00o (200), 64,59o (220), 77,32o (311) dengan ukuran kristal rata-rata 8,7627 nm dan struktur Face Centered Cubic (FCC). Spektrum FTIR ekstrak daun putat dan nanopartikel perak memperlihatkan hilangnya serapan C=O aldehid pada 1736 cm-1 menjadi serapan C=O ion karboksilat pada 1614,42 cm-1 yang menunjukkan adanya reduksi ion perak. Selain itu beberapa serapan puncak lain pada FTIR cenderung berperan sebagai suatu capping agent. Pengujian aktivitas antibakteri nanopartikel perak menunjukkan kemampuan antibakteri yang cenderung sama yaitu kategori sedang terhadap bakteri Escherichia coli dan Staphylococcus aureus dengan diameter zona bening masing-masing sebesar 10,29 mm dan 10,03 mm.
