Abstract

The solubility of medicinal materials is one of the important parameters that will affect the absorption and bioavailability of drugs in the body. Azithromycin belongs to bcs class II which has a low solubility in water. The low solubility of azithromycin causes only a small amount of glibenclamide to be absorbed through the gastrointestinal tract. One solution to improve the solubility of azithromycin by being formulated into a solid lipid nanoparticles (SLN). The manufacturing of azithromycin SLN is carried out by the method of high-speed stirring and ultrasonication. The SLN formula of azithromycin consists of stearic acid, tween 80, PEG 400, PVA, and azithromycin which are varied in number to 0.1 g (F1), 0.2 g (F2), and 0.3 g (F3). The best formula will be determined from the value of the percent EE (%EE). The results of the %EE test of 3 consecutive formulas were 89,073 ± 0.0449%, 86,960 ± 0.0505, and 86,946 ± 0.0554. The results of the heating-cooling cycle stability test on the three formulas obtained an increase in pH in each cycle, the results of the preparation were slightly turbid white, there was a precipitate, and in F2 and F3 there was a phase separation. Mechanical stability tests were obtained in all three formulas. The best formula obtained in F1 with an azithromycin amount of 0.1 g which is further carried out particle characterization and solubility test. Particle characterization results show an average particle diameter of 957.5 nm, PDI of 0.422 ±, and potential zeta 10,2 mV. The results of the solubility test showed that the SLN of azithromycin was soluble in aquadest solvents, 5% HCl, SGF, and SIF. While the NaOH 5% and NaHCO3 5% have a slightly insoluble solubility in the medium. The results showed that azithromycin can be formulated into a soluble SLN.
