Abstract

Striped snakehead (Channa striata) and ocellated snakehead (Channa pleuropthalma) are the species from the genus Channa. The genus Channa consisting of 34 species spread throughout Asia. This research aims to identify the sequence of cytochrome c oxidase subunit I gene DNA mitochondria (COI mtDNA) and to construct the phylogenetic trees among snakehead species. The research methods used in barcoding species and sequences variation were : DNA isolation, amplification by PCR (Polymerase Chain Reaction) and fragmen COI mtDNA sequencing from the fish sample that were collected from Kelekar River, Indralaya, Ogan Ilir Regency. The COI gene were sequenced with length of nucleotide was 604 bp for striped snakehead and 587 bp for ocellated snakehead. Optimum annealing temperature was 500C for 15 seconds with 30 cycles. The result of BLAST analysis showed that striped snakehead from Kelekar River had 99% identity of striped snakehead from Lampung and furthest (96%) with striped snakehead from India. Ocellated snakehead from Kelekar River had 100% similarity with ocellated snakehead from Musi Banyuasin and furthest (83%) with Channa limbata from Myanmar . Water quality measured in Kelekar River were temperature 31,0-31,60C, pH 4.76-4.96, dissolved oxygen 2,7-3,0 mg/L, ammonia <0,009 mg/L, total alkalinity 20 mg/L, and brightness 62,5-63,0 cm.
