Abstract

Enzim azoreduktase diperoleh dari bakteri Bacillus tropicus BD 01. Produksi enzim azoreduktase ditingkatkan dengan cara penambahan inducer. Inducer yang ditambahkan yaitu CaCl2, CaCO3, dan metil merah dengan konsentrasi 1 mM pada saat inokulasi dan kulturisasi. Bakteri Bacillus tropicus BD 01 yang telah diinokulasi dan dikulturkan pada media cair nutrient broth selanjutnya di sentrifugasi dengan kecepatan 3000 rpm selama 15 menit. Ekstrak kasar enzim kemudian di fraksinasi dengan amonium sulfat, diuji aktivitas enzim, ditentukan kadar protein dan dihitung aktivitas spesifik. Hasil aktivitas spesifik enzim azoreduktase ekstraseluler ekstrak kasar tanpa penambahan inducer diperoleh sebesar 0,0029 U/mg. Hasil fraksinasi amonium sulfat pada ekstrak kasar enzim tanpa penambahan inducer memiliki aktivitas spesifik tertinggi pada fraksi 3 dengan tingkat kejenuhan amonium sulfat 40-60% dengan aktivitas sebesar 0,0592 U/mg. Aktivitas enzim azoreduktase ekstraseluler dengan penambahan inducer juga memiliki aktivitas tertinggi pada fraksi ke 3 (40-60%). Inducer yang memberikan aktivitas tertinggi adalah CaCl2, dengan aktivitas spesifik sebesar 0,1440 U/mg, sedangkan penambahan inducer CaCO3 sebesar 0,1159 U/mg dan penambahan inducer metil merah sebesar 0,0659 U/mg.
