Abstract

Alzheimer’s disease is neurodegenerative disease and progressive dementia which is has closely related to age. Etiology of Alzheimer’s disease occurs because excessive production of acetylcholinestrase enzymes in order escape the concentration of acetylcholine and loss of cholinergic markers. One of the therapies than can be used for treatment of Alzheimer’s disease are acetylcholinesterase (AChE) enzyme inhibitor drugs such as donepezil and galanthamine. Difficult availability and high prices of current Alzheimer’s drug therapy make we need for alternatives therapy in the treatment of Alzheimer disease. Natural antioxidant compound are known to reduce or kill neuronal deaths that occur in the pathophysiology of Alzheimer disease due to oxidative stress. Gambir (Uncaria gambir Roxb.) plant has a high antioxidant content such as phenolic and cathechin. Therefore, this study aims to determine the characterization, amount of cathechin content, phenolic compound, IC50 value, and percent inhibiton of gambir water extract on acetylcholinesterase (AChE) enzyme as an agent for Alzheimer’s therapy. Amount of cathechin content, phenolic, flavonoid, IC50 value, and percent inhibition of acetylcholinesterase (AChE) enzyme obtained in this test were 68.1504 0.8868%, 312.8 0.0667 mgGAE/ml, 263.6364 0.3249 mgQE/ml, 54.4090 g/ml dan 34.5912 1.1941%. Quercetin and donepezil were as comparative samples, that strong antioxidant have IC50 value of 2.2406 g/ml and 4.5159 g/ml. Result were analyzed in this test to show gambir water extract classified as first quality, has lower antioxidant activity than quercetin and donepezil, and can inhibition of acetylcholinesterase enzyme (AChE).
