Abstract

This study aimed to determine the total sugar content and antioxidant activity of crude and pure polysaccharide extracts from water lettuce leaves (Pistia stratiotes). In this study, identification of the constituent components of polysaccharides have been done using FT-IR. The research method was carried out experimentally in the laboratory by carrying out crude extraction and purified extraction using the hot air extraction method, which respectively were carried out each 6 times. The data obtained were analyzed descriptively and conducted a different test (Independent Sample t-Test). The test parameters included extract yield, total sugar content test and identification of FT-IR compounds as well as analysis of antioxidant activity. The test results showed no significant difference (p<0.05) to the yield of crude extract polysaccharides and defatted polysaccharides. However, total sugar content test had carried out on crude polysaccharide extract has a higher total sugar content of 96.80±14.60 mg glucose eq./g dry sample rather than defatted polysaccharides of 44.0±2.16 mg glucose eq./g dry sample and antioxidant activity in crude extract of polysaccharides got a higher IC50 value with defatted polysaccharides of 0.484±0.028 mg/mL and 0.406±0.011 mg/mL. Identification of functional groups of polysaccharide compounds in crude extract of polysaccharides and defatted polysaccharides contained functional groups O-H, C-H, and C=O and C-O.
