Abstract

Background: Insomnia is a sleep disorder that adversely affects health and quality of life. Therapy using GABA-A receptor agonists has a variety of side effects. Cinnamon contains anti-inflammatory and antioxidant compounds. This study aims to determine the effect of cinnamon nonpolar fraction (CNF) on GABA levels. Methods: This study is an experimental study in vivo post-test only with a control group. Thirty male Wistar rats were divided into six treatment groups, namely the normal control group, negative control (PCPA + Na-CMC 0.5%), positive control (PCPA + estazolam 0.1 mg/kgBW), and those who received CNF doses of 5 mg/ kgBW, 10 mg/kgBW, and 20 mg/kgBW. CNF was obtained through a separation funnel method with n-hexane solvent from cinnamon simplicia that macerated with 96% ethanol. Induction of insomnia in rats was carried out by administration of p-chlorophenylalanine (PCPA) 300 mg/kgBW via intraperitoneal injection for two days. Administration of CNF and Na-CMC 0.5% was performed with intragastric sonde for 14 days. After that, the rats were euthanized and their hypothalamic tissue was taken to measure GABA levels with ELISA. All statistical analyses, whether univariate, bivariate, or univariate, were performed using IBM SPSS Statistics 26. Results: All CNF groups had higher GABA levels than negative controls. The increase in GABA levels in all groups given CNF was statistically meaningful (p val <0.05). CNF doses of 10 mg/kgBW and 20 mg/kgBW had a better effect in increasing GABA levels of test animals compared to estazolam 0.1 mg/kgBW. Conclusion: CNF had an effect in increasing GABA levels in the hypothalamus of white rats in a model of insomnia. Keywords: nonpolar fraction, Cinnamomum burmanii, insomnia, GABA
