Abstract

Shrimp is a leading commodity for fishery exports, but in recent years production has decreased. One of the factors allegedly causing the decline in shrimp production is due to White Spot Syndrome Virus (WSSV) that can infect vaname shrimp. Cases of WSSV infection need to be detected quickly because it spreads very quickly and can result in mortality up to 100%. The detection technique using Polymerase Chain Reaction (PCR) is one of the ideal techniques in detecting pathogens because the results obtained are very fast and sensitive. This study aimed to detect WSSV disease clinically and molecularly with PCR technology in vaname. This research was conducted in laboratory scale experiments using PCR technique which consisted of sampling, DNA extraction, DNA amplification using PCR, and electrophoresis. Parameters observed were clinical symptoms, DNA band size and water quality. There was no clinical symptoms of shrimp infected with WSSV in this study. Detection of WSSV using PCR found that there was one sample of shrimp from Sungsang which was detected with the WSSV virus with the position of the DNA band showing at 311 and 523 bp. The water quality measurements obtained Dissolved Oxygen (DO) 2.8- 4.2 mg L-1, temperature 28-30.8 oC, salinity 5-29 ppt and pH 7.4-8.3.
