Abstract

Insomnia is a body disorder characterized by decreased sleep quality in the form of difficulty initiating or maintaining sleep. Insomnia is often associated with serotonergic cell death. Cinnamon (Cinnamomum burmannii) is a plant studied to have anti-inflammatory and antioxidant effects. This study aims to determine the efficacy of the nonpolar fraction of cinnamon on serotonin levels. This study is an in vivo experimental study using a post-test only design and a control group at Eureka Research Laboratory. Thirty Wistar male white rats were divided into six groups: normal control, negative control (PCPA + Na-CMC 0.5%), positive control (estazolam 0.1 mg/kgBW), Cinnamon Nonpolar Fraction (CNF) 5 mg/kgBW, CNF 10 mg/kgBW, and CNF 20 mg/kgBW. CNF was obtained from cinnamon simplicia processed through the maceration with 96% ethanol, then continued fractionation using the separating funnel method with n-hexane solvent. Each group induced insomnia, except for the normal control group, by injecting PCPA 300 mg/kgBW intraperitoneally for two days. Na-CMC 0.5% and CNF were administered through an intragastric tube for 14 days. Rats were euthanized and subjected to sagittal decapitation of the brain, and hypothalamus tissue was taken to measure serotonin levels by enzyme-linked immunosorbent assay (ELISA). The mean serotonin levels sequentially in ng/ml are 6,720 ± 0.148; 3,580 ± 0.130; 6,500 ± 0.100; 7,520 ± 0.084; 7,980 ± 0.084; and 8,200 ± 0.122. FNKM 5 mg/kgBW, 10 mg/kgBW, and 20 mg/kgBW can potentially increase hypothalamic serotonin levels compared to normal control, negative control, and positive control groups (p<0.05). CNF has efficacy in restoring hypothalamic serotonin levels in white rat insomnia models. Keywords : Cinnamomum burmannii, Nonpolar fraction, Insomnia, Serotonin.
