Abstract

Latar Belakang: Daun tanaman karamunting memiliki potensi sebagai alternatif terapi diabetes melitus tipe 2 karena mengandung senyawa bioaktif yang memiliki aktivitas antioksidan dan antinflamasi. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian fraksi air daun karamunting (Rhodomyrtus tomentosa) terhadap sekresi insulin pada tikus model diabetes melitus tipe 2 Metode: Penelitian ini menggunakan desain pretest posttest with control group. Penelitian ini menggunakan sampel berupa 25 tikus yang diinduksi dengan diet tinggi lemak dan fruktosa serta injeksi streptozotocin dosis 35 mg/kgBB. Tikus dibagi dalam 5 kelompok yang terdiri atas kelompok perlakuan dengan fraksi air daun karamunting dosis 100 mg/kgBB, 200 mg/kgBB, dan 400 mg/kgBB, kontrol positif (pioglitazone dosis 10 mg/kgBB) dan kontrol negatif. Pengukuran glukosa darah menggunakan metode GOD-PAP dan kadar insulin menggunakan metode ELISA. Penilaian fungsi sekresi insulin menggunakan nilai HOMA-β Hasil: Hasil penelitian menunjukan penurunan bermakna nilai HOMA-β setelah pemberian fraksi air daun karamunting selama dua minggu. Terdapat perbedaan bermakna kelompok perlakuan fraksi air daun karamunting dengan kontrol negatif. Tidak ada perbedaan bermakna nilai HOMA-β antara seluruh kelompok perlakuan fraksi air daun karamunting dengan kelompok kontrol positif pioglitazone. Kesimpulan: Fraksi air daun karamunting tidak berpengaruh terhadap sekresi insulin berdasarkan nilai HOMA-β tikus model diabetes melitus tipe 2.
