Abstract

Oil palm empty fruit bunches (OPEFB) contain high enough cellulose, which is around 49.95%. This study aims to isolate cellulolytic bacteria from oil palm empty fruit bunches and to screen bacteria that have the potential to produce extracellular cellulase using the clear zone method. This research used descriptive explorative and quantitative research methods. The results of the data obtained are presented in tabular form and discussed descriptively. Parameters observed included morphological observations of cellulolytic bacteria isolates, catalase test, motility test, H2S test and Gram staining. This study used selective media Carboxy Methyl Cellulose Bacto Agar 1% (CMC Bacto agar 1%) for bacterial isolation and continued with bacterial screening using 0.1% congo red solution. The results of the study obtained 20 isolates of cellulolytic bacteria and five isolates were selected which had the highest cellulolytic index (IS) value. The cellulolytic index was obtained from the clear zone formed on CMC media. Two bacterial isolates came from the bottom of the OPEFB pile as high as 4 meters in plantations around the factory with IS values of 8.92 and 3.29 at 10-6 dilution. The other two isolates came from the bottom of the OPEFB pile 4 meters high at the factory site at dilutions 10-6 and 10-7 with IS values of 5.27 and 3.35 respectively. Meanwhile, one other isolate at 10-7 dilution with an IS value of 4.02 was found in the middle of the OPEFB pile at the location of the oil palm plantations around the factory. The five selected bacterial isolates dominantly had a round shape but there was one isolate that had an irregular shape. Most of the bacterial isolates were white except for the isolate samples at dilution 10-6 which were yellow in color obtained at the bottom of the OPEFB pile from the location around the factory. Based on the results of Gram staining, it showed the presence of Gram positive bacteria, except for isolation from the bottom of the OPEFB pile at the factory site at dilutions 10-6 and 10-7 which showed Gram negative with bacilli shaped cells. While the isolate samples from the middle and bottom of the OPEFB pile at the oil palm plantation site around the factory was in the form of a coccus.
