Abstract

Senyawa flavonoid antosianin pada kulit buah kopi robusta mempunyai aktivitas antioksidan yang tinggi. Penelitian ini bertujuan untuk mengetahui hasil proses optimasi ekstrak kulit buah kopi robusta menggunakan full factorial design serta karakteristik ekstrak optimal. Metode ekstraksi yang digunakan berupa ultrasound-Assisted Extraction (UAE) dan dianalisis dengan full factorial design 23 (FFD) dengan faktor optimalisasi berupa waktu ekstraksi (10-45 menit), suhu ekstraksi (25-50°C) dan pH pelarut (2-5). Respon yang diamati pada penelitian kali ini berupa % rendemen, total flavonoid, kadar antosianin, dan aktivitas antioksidan. Kondisi optimum dipilih dari kondisi dengan nilai desirability maksimum. Hasil kondisi optimum didapatkan pada waktu ekstraksi 45 menit, suhu ekstraksi 50°C dan pH 2 dengan hasil respon rendemen 17,25 %, TFC 242,259 mgCE/gr, Antosianin 367,041 mg/100 gr antioksidan 22,105ppm dan nilai desirability 0,999. Karakteristik ekstrak dengan kondisi optimum memiliki kadar air 0,21% dan susut pengeringan 0,26%. Hasil skrining fitokimia ekstrak kondisi optimum didapatkan positif mengandung fenolik, flavonoid, tanin, saponin dan alkaloid. Hasil verifikasi ekstrak optimum didapatkan nilai rendemen 17,10%, TFC 250,282 mgCE/gr, antosianin 391,199 mg/100gr, dan aktivitas antioksidan 24,539 ppm. Hasil respon menunjukkan bahwa nilai berada pada kisaran 95% CI dan 95% TI sehingga nilai prediksi mendukung verifikasi.
