Abstract

Flaxseed oil (Linum usitatissimum) has been shown to enhance the function of the antioxidant system due to its antioxidant compounds such as phenols and vitamin E. The development of flaxseed oil into stable dosage forms such as nanoemulsions is very potential from the point of view of the efficacies contained. This research was conducted to determine the concentration of flaxseed oil in nanoemulsion dosage which had the best antioxidant activity and then analyzed with SPSS®23 Shapiro-Wilk. Flaxseed oil was formulated into a nanoemulsion dosage form using various concentrations of 5%, 8%, 10%, and 15% flaxseed oil by adding Tween 80 with a concentration of 40% as a surfactant and Sorbitol with a concentration of 20% as a cosurfactant. The physical evaluation of nanoemulsion preparations included organoleptic, homogeneity, pH, percent transmittance, specific gravity, viscosity, type of nanoemulsion and physical stability cycling test. Then the antioxidant activity test was carried out to determine IC50 using the DPPH method. According to the analysis results of Shapiro-Wilk's SPSS®23 program, it can be stated that the oil concentration of 5% and 8% has a significant effect on determining the best formula produced. Based on the research results obtained, the best formula is formula 1 and 2 with an IC50 of 2,367 µg/mL and 2,305 μg/mL due to the highest antioxidant activity in the very strong category and when compared to Vitamin C as a positive control which has an IC50 value of 2,321 μg/mL not significantly different. Keywords : Antioxidant, DPPH, Flavonoid, Flaxseed Oil (Linum usitatissimum), Nanoemulsion, Sorbitol, Tween 80 Minyak biji rami (Linum usitatissimum) telah terbukti meningkatkan fungsi sistem antioksidan karena senyawa antioksidan nya seperti fenol dan vitamin E. Pengembangan minyak biji rami menjadi bentuk sediaan stabil seperti nanoemulsi menjadi sangat potensial jika terkait dengan banyaknya khasiat yang dimiliki. Tujuan penelitian ini untuk menentukan konsentrasi minyak biji rami dalam sediaan nanoemulsi yang memiliki aktivitas antioksidan terbaik kemudian dianalisis dengan SPSS®23 Shapiro-Wilk. Minyak biji rami diformulasikan menjadi bentuk sediaan nanoemulsi dengan menggunakan variasi konsentrasi minyak biji rami 5%, 8%, 10%, dan 15% dengan menambahkan Tween 80 dengan konsentrasi 40% sebagai surfaktan dan Sorbitol dengan konsentrasi 20% sebagai kosurfaktan. Evaluasi fisik sediaan nanoemulsi meliputi organoleptis, homogenitas, pH, persen transmitan, bobot jenis, viskositas, tipe nanoemulsi dan stabilitas fisik cycling test. Kemudian uji aktivitas antioksidan penentuan IC50 dengan metode DPPH. Menurut hasil analisis program SPSS®23 Shapiro-Wilk yang didapatkan maka dapat dinyatakan bahwa konsentrasi minyak 5% dan 8% berpengaruh signifikan terhadap penentuan formula terbaik yang dihasilkan. Berdasarkan hasil penelitian yang didapatkan, formula terbaik merupakan formula 1 dan 2 dengan IC50 sebesar 2,367 µg/mL dan 2,305 µg/mL dikarenakan aktivitas antioksidan paling tinggi dengan kategori sangat kuat serta dibandingkan Vitamin C sebagai kontrol positif yang mempunyai nilai IC50 sebesar 2,321 μg/mL tidak berbeda signifikan.
