Abstract

Synthetic dye waste direct red 80 from the textile industry and textile products is very difficult degrdated in nature due to its complex structure and highly toxic nature. Several microorganisms, one of which is bacteria, are known to have the ability to biodecolorize synthetic dyes. Further exploration of thermophilic bacteria originating from hot springs, especially from Semendo, needs to be carried out in order to obtain bacteria that have the potential to be biodecolorizing agents for synthetic dyes. This research was conducted from August 2022 to January 2023 at the Genetics and Biotechnology Laboratory, Microbiology Laboratory, Physiology and Development Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University, Indralaya. The stages of the research consisted of rejuvenation, selection of thermophilic bacteria to decolorize direct red 80, measurement of biodecolorization ability, optimization (pH, temperature, and agitation speed), analysis of biodecolorized products, characterization, and genotypic identification using the 16S rRNA gene barcode. All bacterial isolates from Semendo hot springs were able to decolorize direct red 80 at a concentration of 80 ppm for 5 days with the highest of biodecolorizing ability owned by SAA isolates of 59.82 ± 0.89%. The optimum percentage of biodecolorization power of SAA isolates was 66.94 ± 0.48% at pH 9, temperature 75oC, with an agitation speed of 120 rpm. The results of the FT-IR spectrum analysis indicated the detection of amine groups that can be evaluated by the formation of peaks at 1635.00 cm-1, 1154.13 cm-1, and 1076.31 cm-1. The DNA sequence of Isolate SAA has high similarity with Paenibacillus phoenicis strain 3PO2SA, which is 98.55%.
