Abstract

ANTAGONISM ABILITY ENDHOPHYTIC FUNGI OF KABAU (Archidendron bubalinum (Jack) I.C. Nielsen) FRUIT AGAINST Colletrichum capsici IPBCC 13.1098 Anggi Sandra 08041381924091 ABSTRACT One of the obstacles in the cultivation of red chili is anthracnose disease or fruit rot which has been widely reported as one of the diseases that often attacks red chili cultivation. Efforts to control and prevent Colletotrichum capsici the cause of anthracnose disease usually use synthetic pesticides, due to some of the adverse effects that arise due to the use of pest and disease control chemicals, alternatives that can be used besides reducing side effects and saving costs by using biological control agents using several microorganisms. This research is important for microscopic observation using a Scanning Electron Microscope (SEM) to determine the potential of the endophytic fungus of kabau fruit (Archidendron bubalinum (Jack) I.C. Nielsen ) as a biocontrol agent Colletotrichum capsici that causes anthracnose. An alternative is needed by using kabau fruit (Archidendron bubalinum (Jack) L.C. Nielsen) as endophytic fungi. This research was conducted from 1 September 2022 to 1 February 2023 at the Microbiology Laboratory, Genetics and Biotechnology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University, Indralaya. The stages of the research consisted of making media, sterilizing material tools, propagating endophytic fungi, antagonistic test of kabau fruit endophytic fungi, inhibition proportions, observing microscopic mechanisms using a light microscope and Scanning Electrone Microscope (SEM), characterization of kabau fruit endophytic fungi isolates and assistance. Seven isolates of kabau fruit endophytic fungus were found which had the ability to inhibit the growth of Colletotrichum capsici IPBCC 13.1098 which had different proportions of inhibition ranging from 8.33% - 70.58%, 1 isolate of weak inhibition, 5 isolates of moderate inhibition and 1 isolate of moderate inhibition. strong. As a result of the antagonistic activity of the endophytic fungi of the kabau fruit, the occurrence of defense mechanisms such as competition, antibiosis and mycoparasitism also results in abnormalities in the shape of the hyphae, such as lysis, swelling of the hyphae and hyphae twisting. KBM1J1 isolates were identified as Botrytis, BKT1J1 isolates were identified as Culvularia and KBT1J1 isolates were identified as Fusarium.
