Abstract

Peat forests in Indonesia have a diversity of flora and fauna that have unique biodiversity. One of the plant genera found in peat forest ecosystems is the Tristaniopsis genus. Pelawan Putih (Tristania whitiana Griff.) plant has been widely used by the community as a traditional medicine for digestive ailments such as diarrhea because it is rich in essential oils which are used as antibacterial, antioxidant and anti-fungal drugs. Antioxidant chemical compounds contained by fighters are flavonoids, terpenoids, tannins and saponins. This study aims to determine the antioxidant activity of the fraction, compound class and IC50. This research was conducted from August 2022 to December 2022 at the Biology Department Laboratory, FMIPA, Sriwijaya University. The research procedure was started with sample preparation, followed by extraction, fractionation using the liquid-liquid method, then purified and isolated by column chromatography and tested for the antioxidant activity of the compound using the DPPH (1,1-diphenyl-2-picrylhydrazyl) method. The results obtained from this study were that in the extraction process, 154.4 grams of methanol viscous extract was obtained with a weight percent yield of 20.58%. The results of pelawan putih leaf fractionation obtained the percent yield of n-hexane, ethyl acetate and water methanol fractions, namely 6.52%, 49.48% and 43.98%, respectively. The results of the antioxidant activity test of Pelawan leaves used a TLC plate of the n-hexane and ethyl acetate fractions. Pure isolates N1, N2, E1, and E2 belong to the class of terpenoid compounds with IC50 values of 20.72 ppm, 16.68 ppm, 9.91 ppm and 46.01 ppm, indicating strong antioxidant activity. E3 is included in the class of terpenoid compounds but has very weak antioxidant activity with a value of 368.05 ppm. Pure isolates N3 and N4 belong to the group of steroid compounds that have strong antioxidant activity as indicated by IC50 values of 28.04 ppm and 40.62 ppm respectively. Pure isolates containing terpenoid compounds, namely N4 with an IC50 of 111.32 indicating moderate antioxidant activity and E6 with an IC50 of 79.64 indicating strong antioxidant activity.
