Abstract

Daun sambiloto (Andrographis paniculata (Burm f.) Wall. ex Nees) mengandung senyawa terpenoid utama berupa andrografolid dan senyawa flavonoid berupa kuersetin yang memiliki potensi sebagai agen fotoprotektif. Pembuatan nanoemulsi ekstrak daun sambiloto dilakukan untuk meningkatkan bioavailabilitas, tampilan serta stabilitas ekstrak. Penelitian ini bertujuan untuk mengetahui konsentrasi optimum tween 20 dan PEG-400 dalam sediaan nanoemulsi ekstrak daun sambiloto menggunakan metode desain faktorial 2² piranti lunak Design Expert seri 12® dengan respon berupa nilai pH, densitas, viskositas dan persen transmitan. Formula optimum yang dihasilkan dilakukan uji PSA, uji aktivitas fotoprotektif berupa penentuan nilai sun protection factor (SPF) dan uji antioksidan, serta uji stabilitas. Ekstrak daun sambiloto memiliki kadar andrografolid 3,397% dan flavonoid 2,868%. Formula optimum sediaan nanoemulsi diperoleh pada konsentrasi tween 20 10% dan PEG-400 10%. Hasil uji PSA sediaan nanoemulsi memiliki ukuran globul 130,43 ± 54,056 nm, nilai PDI 0,318 ± 0,043, dan zeta potensial -26,5 ± 0,544. Pengujian aktivitas fotoprotektif formula optimum memperoleh nilai SPF 42,444 ± 0,021 dan nilai IC₅₀ 103.791± 1.234. Uji stabilitas cycling test tidak mengalami perubahan yang signifikan (p<0,05) dan uji sentrifugasi tidak ditemukannya adanya perubahan fisik. Berdasarkan hasil yang diperoleh formula optimum sediaan nanoemulsi ekstrak daun sambiloto memiliki ukuran nano yang tergolong polidispersi dengan potensi SPF tinggi dan antioksidan sedang dan stabilitasnya cukup baik.
