Abstract

Tilapia merupakan ikan introduksi untuk akuakultur dan terus mengalami pengembangan melalui hibridisasi. Tujuan penelitian ini adalah untuk mengetahui sekuen COI DNA mitokondria, jarak genetik dan filogenetik. Penelitian ini dilaksanakan pada bulan September 2022-Januari 2023. Sampel didapatkan dari Danau Toba, Danau Ranau dan Balai Riset Pemuliaan Ikan (BRPI) Sukamandi Jawa Barat. Sampel diambil sebanyak lima ekor dari setiap danau dan masing�masing lima ekor strain ikan nila biru dan nila merah hasil pengembangan genetik di BRPI. Tahapan DNA barcoding terdiri dari ekstraksi DNA, amplifikasi DNA menggunakan Polymerase Chain Reaction (PCR), elektroforesis, dan sekuensing gen COI. Fragmen gen COI mtDNA diamplifikasi pada suhu 52 oC selama 45 detik dengan 31 kali siklus. Panjang nukleotida gen COI sampel 690 base pairs (bp). Hasil Basic Local Alignment Search Tool nucleotide (BLASTn) sampel TTA2, TTB1, TTB2, TTB3, TTB4, TTB5, TRA3, TRA4, TRB4, TRR1, TRR3, TRR5, TBR1, TBR2, TBR3, TBR4 dan TBR5 berkerabat dengan ikan nila Nigeria dan Malaysia (99,42-100%). Sampel TTA1, TTA3, TTA4, TRA5 dan TRB3 berkerabat dengan ikan mujair Filipina (99,42-100%) dan Thailand (99,42%). Sampel TRA1 berkerabat dengan ikan nila biru Nigeria (100%). Sedangkan TTA5 berkerabat dengan ikan nila biru Filipina (100%), China (99,27%) dan Nigeria (99,70%). Sampel TRB1, TRB2, TRB5 dan TRR4 berkerabat dengan spesies Oreochromis urolepis Malaysia (99,42-100%) dan Israel (100%). Seluruh sampel memiliki jarak genetik sebesar 0,00 (0%) terhadap masing-masing spesies hasil BLASTn. Pohon filogenetik sampel dari masing-masing lokasi membentuk empat subcluster terpisah (O. niloticus, O. mossambicus, O. aureus, O. urolepis) namun masih dalam satu cluster yang sama dengan nilai bootstrap sebesar 90%. Hasil pengukuran parameter kualitas air meliputi suhu yang berkisar antara 24,6-30,3oC, kecerahan perairan 0,35-5,60 m, pH 7,0-8,5, DO 3,82-5,8 mgL-1 , amonia 0,07-0,21 mgL-1 , alkalinitas 38- 60 mgL-1 .
