Abstract

Test has been carried out isolation and enzyme activity of soybean lipoxygenase. Fat-free soy bean flour is made by soaking using chloroform methanol solvent to produce a fat-free soy flour amounted to 87,17%. Lipoxygenase enzyme was isolated by extraction using distilled water so that the lipoxygenase enzyme extracts obtained by 16 mL/g. Enzyme activity assay using methods Hamberg & Samuelsson in order to obtain lipoxygenase enzyme activity of 1460 Units/mL. The protein content is measured by the enzyme extract biuret method and obtained the protein content of 2,72 mg/mL. Subsequently extract characterized enzymes and enzymes get optimum conditions at the time of 5 minutes, the temperature 25 degrees celcius, the concentration of the substrate 2 mL and pH 9 with activity of 3428 units/mL.
