Abstract

Papaya leaf (Carica papaya L.) is estimated to have ability as a natural larvacide material for controlling Aedes aegypti mosquito that causes Dengue Fever (DBD). The alkaloids, flavonoids, samponins, and tanins contents has a larvacidal activity. The papaya leaf is extraction in this study was formulated in microparticle preparations. The microparticle preparations of papaya leaf extract consist of chitosan polymer, sodium alginate, and cross linker calcium chloride by using ionic gelation method. While, the variations of calcium chloride concentration is conducted on three formulas yielding percent of entrapment efficiency (EE) of 99.33% (20 μL), 99.48% (40 μL), 99.35% (100 μL), respetively. Based on the results of EE percent, it‟s obtained the microparticles optimum formula of papaya leaf extract using chitosan polymer sodium alginate with a concentration of calcium chloride of 40 μL. The result of microparticle characterization such as diameter and particle distribution (poly dispersity index) using particle size analyzer (PSA) tool in the optimum formula is 1269.9 nm and 0.434. Particle morphology observation using transmission electron microsope (TEM) in microparticle formula of papaya leaf extract showed a particle whit spheric shape. This study was conducted with 8 treatments and 2 controls, each consisting of 20 larva of Aedes aegypti instar III. The observation time is 24 hours. The activity test of papaya leaf extract was carried out with concentration variations of 200, 500, 800, 1100, 1400, 1700, 2000, and 2300 ppm. While, the microparticle activity test of papaya leaf extract was carried out with variations of 75, 100, 125, 150, 175, 200, 225, and 250 ppm. In the result of probit analysis with linear regression, it‟s obtained LC50 papaya leaf extract amounted to 1059 ppm whereas in micoparticle of papaya leaf extract, it‟s obtained LC50 amounted to 125.054 ppm. The result os statistical analysis using Kruskal Wallis an Mann-Whitney test showed that all concentration in treatment group had significant difference on the ontrol group with p < 0.05.
