Abstract

Liquid waste from textile industry is not only about concentrated dye but also complex organic compound that hard to degradate such as lignin and cellulose. Lignin and cellulose that blend with dye can make the liquid waste hard to degradate naturally, thus a further decoloration is needed. Biological decoloration using themophilic bacteria is more effective than physically dan chemically. The using of thermophilic bacteria can make decoloration perform faster, decrease contamination, lower cost, and eco-friendly product. The ability of thermophilic bacteria isolates from Tanjung Sakti hot spring to degradate lignin and cellulose is unknown so far, thus screening is needed. Thermophilic bacteria isolates that can degradate lignin and cellulose will identified using 16S rRNA gene fragment to discover the species. This research was conducted from November 2016 until February in Laboratory of Genetics and Biotechnology, Biology Department, Faculty of Mathematics and Natural Science, Sriwijaya University. This research was aimed to get thermo-lignocellulolytic bacteria from Tanjung Sakti hot spring bacteria isolates and also to identify the species and genetic relationship from thermo-lignocellulolytic bacteria using 16S rRNA gene. This research started by screening thermo-lignocellulolytic bacteria isolates using mineral agar medium enriched with methylen blue 25 mg/L and carboxymethyl cellulose (CMC) 1% volume, isolation of bacteria DNA, 16S rRNA gene amplification using Polymerase Chain Reaction method, agarose electrophoresis, sequencing by South Korea Macrogen, 16S rRNA fragment sequence analysis using BLAST method by https://blast.ncbi.nlm.nih.gov/Blast.cgi., and phylogenetic tree construction using MEGA6. The result of this research are data of thermolignocellulolytic bacteria, electroforegram of isolated DNA and amplificated 16S rRNA fragment, and also phylogenetic tree. The conclusion of this research, there are five thermo-lignocellulolytic bacteria isolates, those are TS 3, TS 4, TS 8, TS 12, and TS 16. Identification result showed that each percentage of TS 3, TS 8, and TS 16 identified as Bacillus licheniformis strain DSM 13 are 93%, 95%, and 92%, TS 4 isolate identified 94% as Anoxybacillus rupiensis strain R270, and TS 12 isolate has 93% identity pencentage as Aeribacillus pallidus strain DSM 3670.
