Abstract

Antioxidant has a molecular compound structure that can give electrons to free radical molecules and can break the chain reaction of free radicals. The antioxidant can slow or inhibit the oxidation of the sustances which easily oxidized in low concentrations. Robusta coffee leaves (Coffea canephora Sp Var. Robusta) contain compounds like flavonoid, phenolic, and tannin morr than arabica coffee leaf. The flavonoids and phenolics substances have ability as antioxidant. This research are to optimize yield result and content of flavonoid total from robusta coffee leaves. Based on the effect of ethanol concentration (30, 70, and 96% v/v) and stirring time (30, 60, and 90 minutes) against flavonoid extraction with factorial experiments. The differences of stirring time and solvent concentration with different polarities affect the total gain of flavonoid and yield value. The total number of flavonoids and the highest yield value become reference as an optimal treatment. Based on analysis design expert, the optimal treatment result in extraction with 30% ethanol solvent concentration and stirring time in 58.983 minutes. This result than performed an antioxidant test with FRAP (ferric reducing ability of plasma) method to measure the antioxidant capacity of the robusta coffee leaves extract. The optimal treatment of robusta coffee leaves extract obtained flavonoid total amount 175.865 mgQE/g and yield value amount 15.044%. That also showed IC50 strength is 96.019 μg/mL.
