Abstract

Endophytic microorganisms are microorganisms that can live in the network (host) plant without damaging the existing network on the plant. One endophytic microorganisms are endophytic fungi are capable of producing bioactive compounds with the potential to be developed into raw materials for drugs, including anticancer and antibiotic. Endophytic fungi can be isolated from plant tissue systems such as seeds, leaves, flowers, twigs, stems and roots. Various functional compounds can be isolated from endophytic fungi. Isolation of endophytic fungi can be made from plants with the potential to be used as raw materials for drugs such as plants Setunjang Langit (Helminthostachys zeylanica (L.) Hook). Currently the plant Setunjang Langit was getting hard to find, so the things you can do to keep preserve plants Setunjang Langit and that the efficacy of this plant can still be used is to utilize secondary metabolites endophytic fungi found in plants Setunjang Langit (Helminthostashys zeylanica (L.) Hook). Secondary metabolites produced from endophytic microorganisms is expected to produce compounds that are antibacterial. The purpose of this study was to obtain a type of fungi endofitik of plants Setunjang Langit, getting secondary metabolites of fungi endofitik plants Setunjang Langit potential as an antibacterial, getting MIC secondary metabolites of fungi endofitik of plants Setunjang Langit that is antibacterial, and determine the characteristics and identification of fungi endofitik plants Setunjang Langit potential as an antibacterial. The research was conducted on August to December 2016, sampling was conducted in the village of Lubuk Sakti Ogan Ilir regency, South Sumatra Province. The process of isolation, selection and characterization of endophytic fungi plants Setunjang Langit take place at the Laboratory of Microbiology, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University. This research was conducted through several phases of work, namely, sampling and sample preparation, isolation, purification, production and extraction of secondary metabolites, anti-bacterial test, testing MIC, Thin Layer Chromatography, macroscopic and microscopic characterization of endophytic fungi, as well as the identification of endophytic fungi. The results obtained in this study were obtained 6 isolates of endophytic fungi on the isolation and purification process includes three isolates on leaves, 2 isolates at the root, and 1 isolate stem. The test results of antibacterial activity obtained secondary metabolites isolates DTLJ4 have antibacterial activity Salmonella thypi and Escherichia coli, secondary metabolites isolates DTLJ5 have antibacterial activity Salmonella thypi, while extracts ATLJ3B, BTLJ2, DTLJ3, and ATLJ1 not have antibacterial activity Salmonella thypi, Shigella dysenteriae , Staphylococcus aureus, and Escherichia coli. MIC extract of secondary metabolites the bacteria Salmonella thypi isolates DTLJ4 1% to produce inhibition zone diameter of 6.07 mm, while the bacterium Escherichia coli at a concentration of 1% extract yield of 6.37 mm zone of inhibition. DTLJ4 fungal isolates have the same characteristics as the species fungi isolates Penicillium brevicompactum while DTLJ5 have the same characteristics as the fungi Aspergillus carbonasius.
