Abstract

Efforts for malaria control and prevention are inhibited by drug and insecticide resistance. Thus, malaria vaccine is urgently needed. Merozoite surface protein 1 Plasmodium falciparum are potential vaccine candidate antigens. However, extensive genetic diversity of these antigens becomes a major difficulties for the development of an effective vaccine. Furthermore, the genetic diversity in malaria parasite population is one of an important indicator of the malaria transmission intensity in an area. The aim of this study was to analyse genetic diversity of MSP-1 genes in P. falciparum isolates from four districts in South Sumatera. A total of 40 P. falciparum infected blood samples from patients with uncomplicated malaria attending four districts (RSUD Baturaja, RSUD Lahat, RSUD Sekayu/Puskesmas Ds. Lubuk Bintialo and RSMH Palembang) were collected between February and May 2014. DNA were isolated and msp-1 genes were genotyped using allele-spesific nested PCR. MSP-1 allelic families were identified in 25 blood samples. A majority of patients had multyclonal infections. The proportion of isolates having only K1 and MAD20 alleles was 0/25 (0%) and 7/25 (28%) respectively. Both allelic families were found in 18/25 (72%) of the isolates. No RO33 alleles were found in this study. The overall multiplicity of infections (MOI) was 3,6. This is the first report on molecular characterization of P. falciparum field isolates in South Sumatera. High level of mixed and multiclonal infections, along with a high degree of MOI, showed an extensive genetic polymorphism of MSP-1 in P. falciparum isolates in South Sumatera.
