Abstract

Vernonia amygdalina is one of the traditional medicinal plants used for the treatment of diabetic. Efficacy as a diabetic is related to antioxidant compounds. The research was conducted to find out the result of comparison between the soxletation and maceration extraction methods on antibacterial and antioxidant activity. This research begins with extraction by maceration and soxhlation using ethanol and ethyl acetate solvents. Each extract is determanined % of yield and water content used the gravimetric method, determination of total phenolic and flavonoid levels using a UV-Vis Spectrophotometer. Antioxidant activity was tested using the DPPH method (1,1-diphenyl-2-picril hydrazil) and antibacterial using diffusion method of against Escherichia coli, Shigella dysenteriae, Staphlococcus aereus, and Bacillus subtilis. The results showed that the V. amydalina extract with ethyl acetate solvent with maceration method had the highest total flavonoid (38,36 mg QE/g) and extract with ethyl acetate solvent with maceration method had the highest total phenolic (21,67 mg GAE / g). The results ethanol extract with maceration method showed higher antioxidant property compared all other extracts with IC50 494,86 mg/L. The evaluation to antibacterial properties The extracts of V. amyigdalina showed as weak antibacterial activity against all bacteria test with inhibition zone 6.1 ± 0.1 to 9.4 ± 0.9 mm at variation concentration 62.5 – 1000 μg/mL
