Abstract

Gambir water leaves extract contains catechin compound which have activities as anti-inflammatory, antibacterial, and antioxidants formulated into preparations of gingival mucoadhesive patch for the therapy of gingivitis. The dosage form formulation used a combination of chitosan and tapioca starch with a factorial design 22. Determination of the optimum formula based on the test results data was analyzed using DX® 10. The composition of chitosan and tapioca starch in the optimum formula was 900 mg and 300 mg with a desirability value of 0.968. The obtained results of the optimum response formula had RSD weight uniformity of 0.4805 ± 0.1887%, RSD thickness uniformity of 0.9716 ± 1.2026%, surface pH 6, folding endurance >300 folds, percent elongation of 53.333 ± 0.1082%, swelling index of 57.7667 ± 1.2050%, and mucoadhesive time 320 ± 1.1547 minutes. Chitosan increased thickness, percent elongation, swelling index, and mucoadhesive time but decreased weight uniformity of patch. Modified tapioca starch increased weight uniformity and mucoadhesive time, but decreased thickness, percent elongation, and swelling index. Catechin content of optimum formula of 92.1667 ± 0.3626%. The results of analysis % diffused the optimum formula and pure extract at 360 minutes by 59.0621 ± 0.0799% and 66.6596 ± 0.0982%. Compartmental analysis was observed using WinSAAM® resulting in pure extract and optimum formula following the 3 compartment model with the correlation between Qo and Qc with p-value <0.05. Study of catechin content stability using heating/cooling method in 3 cycles showed that optimum formula more stable compared pure extract. FTIR test results showed that there was no chemical interaction beetween gambir water leaves extract, polymer, and plasticizer.
