Abstract

Kulit ikan gabus mengandung senyawa kolagen yang berperan dalam fase proliferasi dan maturasi dalam penyembuhan luka bakar. Kolagen kulit ikan gabus diekstraksi dengan metode Acid Soluble Collagen (ASC) menggunakan asam asetat 0,5 M dan memiliki rendemen sebesar 2,321%. Ekstrak kolagen kulit ikan gabus diformulasikan menjadi bentuk sediaan spray gel untuk mengurangi kontaminasi atau infeksi serta rasa sakit ketika diaplikasikan pada bagian luka. Spray gel diformulasikan menggunakan HPMC-60SH dan Carbopol®940 sebagai gelling agent. Kedua gelling agent dioptimasi menggunakkan program Design Expert®10 dengan metode desain faktorial 22. Konsentrasi gelling agent yang dioptimasi sebesar 0,5% sebagai low level dan 2% sebagai high level. Sediaan dievaluasi sifat fisik yang meliputi organoleptis, homogenitas, viskositas, pH, daya lekat, daya tercuci, pola penyemprotan, dan pump delivery. Hasil pengujian dianalisis dengan DX®10 untuk mendapatkan formula optimum. Didapatkan konsentrasi optimum HPMC-60SH 1,733% dan Carbopol®940 1,807% memiliki diameter pola penyemprotan 1,013 ± 0,033 cm, pH 7,067 ± 0,0603, daya lekat pada kulit 24,107 ± 0,067 detik, daya lekat pada kertas saring 24,273 ± 0.103 detik, daya tercuci 6,333 ± 0,289 mL, dan pump delivery 6,447%.
