Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh penambahan asam amino metionin terhadap pertumbuhan dan pembentukan lovastatin oleh bakteri Lactobachilus achidophilus asal bekasam. Penelitian ini menggunakan metode Rancangan Acak Kelompok (RAK) dengan 4 taraf perlakuan. Masing-masing perlakuan dilakukan pengulangan sebanyak 3 kali, dengan ulangan sebagai kelompok. Perlakuan yang diterapkan pada penelitian ini adalah kultur bakteri L. acidophilus menggunakan MRS dan metionin 0% (P1); metionin 0,1% (P2); metionin 0,2% (P3); metionin 0,3% (P4). Parameter yang diamati adalah pertumbuhan bakteri, pengukuran nilai pH dan analisis lovastatin. Hasil uji nilai OD L. achidophilus selama inkubasi cenderung meningkat dari hari ke 2 sampai hari ke 6 dan menurun pada hari ke 8. Puncak pertumbuhan bakteri terjadi pada hari ke 6 dan menurun pada hari ke 8. Uji nilai pH menunjukkan bahwa pH dari perlakuan penambahan metionin secara keseluruhan yang paling rendah terdapat pada hari ke 6 dengan kisaran nilai pH 3,11 - 3,54 dan mengalami peningkatan pada hari ke 8. Perlakuan penambahan berbagai konsentrasi metionin dapat meningkatkan kandungan lovastatin namun tidak berbeda nyata secara statistik.
