Abstract

Common Snakehead is an economic fish with a wide area distribution. Common Snakehead family Channidae has 2 genera, namely Channa (34 species) and Parachanna (3 species). This diversity can be caused by the diversity of nucleotides and amino acids among species. The purpose of this research was to obtain cytochrome c oxidase gene sub unit 1 (CO1) mitochondrial DNA to find out the diversity of nucleotides and amino acids in the COI mtDNA and to know the trees of phylogenetic and genetic distance between Common Snakehead. The methods used in species barcoding and sequence variation were DNA isolation, amplification using PCR (Polymerase Chain Reaction) and sequencing of the COI mtDNA gene region from fish samples obtained from the Kelekar and Batanghari rivers. This research was conducted from January 2019 to February 2020. The length of COI gene of Common Snakehead nucleotides was aprroximately 700bp. BLAST analysis showed Common Snakehead from the Kelekar river and Batanghari river have the closest similarity of 99.5% with Common Snakehead from Singapore MF496938.1 and farthest with Common Snakehead from India KJ847132.1 96.66%. The results of water quality analysis from the Kelekar and Batanghari rivers were temperature 27.8-31.0oC, pH 5.0-7.9, dissolved oxygen 3.80-6.13 mgL-1, and ammonia 0.04-0.10 mgL-1.
