Abstract

Ocellated Snakehead (Channa pleuropthalma) is a fish that belongs to the Channidae fish family. Ocellated Snakehead fish has a special characteristic, orange-spotted found in the body. The purpose of this study is to obtain genetic information of CO1 mt DNA in Ocellated Snakehead fish to support breeding efforts and to know the diversity of nucleotides and amino acids among Ocellated Snakehead and others Channa’s fish based on CO1 mt DNA gene. The result of COI mt DNA gene sequencing showed that the nucleotide of Ocellated Snakehead was 661 base pairs. Analysis of sequences indicated that Ocellated Snakehead from the Kelekar river showed no diversity with Ocellated Snakehead from Banyuasin (IDN), Ontario (CND), Frankfurt (DEU). In comparison to others Channa’s fish there were quite a lot of differences in nucleotides and amino acids that are formed. There were 103 nucleotide differences with 8 changes in amino acids from Striped Snakehead, 84 nucleotide differences with 79 changes in amino acids from C. diplogramma. In comparison to C. gachua there were 104 nucleotide differences with 92 changes in amino acids. There were 105 nucleotide differences with 99 changes in amino acids from C. marulius, 102 nucleotide differences with 94 changes in amino acids from C. melasoma, and 87 nucleotide differences with 84 changes in amino acids from C. micropeltes. The results of measuring the quality of water in the Kelekar river were 29.3-30.70C for temperature, pH 4.6-6.7, dissolved oxygen 1.31-3.76 mg.L-1 and ammonia 0.17-0.20 mg.L-1.
