Abstract

Etosom metilprednisolon ditujukan untuk memperbaiki sistem penghantaran obat dan untuk meningkatkan penetrasi obat dari metilprednisolon. Etosom metilprednisolon dalam penggunaannya memiliki beberapa kekurangan, oleh karena itu maka dibuat ke dalam bentuk gel untuk memudahkan alam penggunaannya dan untuk memperbaiki penghantaran obat dalam tubuh. Formulasi gel etosom metilprednisolon dilakukan dengan mengkombinasikan basis gel karbopol dengan variasi konsentrasi 0,5%, 1% dan 1,5%. Formula terbaik yang didapatkan lalu diujikan dengan pengujian antiinflamasi dengan menggunakan tikus putih jantan galur Wistar yang dikelompokkan menjadi 3 kelompok, yaitu kelompok sampel (Gel etosom metilprednisolon), kelompok positif (Krim metilprednisolon), dan kelompok negatif (Gel etosom tanpa zat aktif metilprednisolon). Metode yang digunakan untuk menguji antiinflamasi adalah menggunakan penginduksi karagenan. Gel etosom metilprednisolon dikarakterisasi dengan cara mengamati organoleptis, homogenitas, pH, daya sebar, daya lekat, daya proteksi, daya tercuci, viskositas dan stabilitas dengan metode centrifugal test serta cycling test. Karakteristik dari formula terbaik gel etosom metilprednisolon menunjukkan hasil yang baik hampir pada keseluruhan uji. Diperoleh formula terbaik untuk gel etosom metilprednisolon dengan konsentrasi karbopol sebesar 1%. Hasil dari uji antiinflamasi menunjukkan bahwa tidak ada perbedaan yang signifikan dalam penurunan udem pada kelompok sampel serta kelompok positif, tetapi terdapat perbedaan yang sangat signifikan pada kelompok negatif. Ethosome methylprednisolone is used for drug delivering system and to enhance the possibility of drug's penetration of methylprednisolone. Ethosome methylprednisolone in usage has several weaknesses, that was why the ethosome was then being made into gel to make it easier to use and for better delivery system in body. The formulation of ethosome methylprednisolone gel was combined by carbopol gel with varying concentration from 0,5%, 1% and 1,5%. The best formula obtained was the 1% variation. The best formula was then being testes for antiinflammation test with Wistar white rat male mixe into 3 divided groups, sample group (ethosome methylprednisolone gel group), positive group (methylprednisolone cream), and negative group (ethosome gel without methylprednisolone). The method used for testing the antiinflammation was carageenan induction. Ethosome methylprednisolone gel is being characterized by observing organoleptic, homogenity, pH, spreadability, clingability, protection power, washability, viscosity, and stability by centrifugal test method and cycling test. The characterization of the best obtained formula showed it was good at every tests. The result of antiinflammation test was that there was no significant difference in udemic group in sample group and positive group, but a lot of significance in negative group.
