Abstract

Daun kayu manis (Cinnamomum burmannii) dan rimpang temulawak (Curcuma xanthorrhiza Roxb) diketahui mengandung minyak atsiri. Penelitian ini bertujuan untuk mengetahui komponen minyak atsiri dan menentukan aktivitas antibakteri kombinasi minyak atsiri daun kayu manis dan rimpang temulawak terhadap mikroba uji Staphylococcus aureus dan Escherichia coli serta Konsentrasi Hambat Minimum (KHM) terhadap masing-masing mikroba uji. Isolasi minyak atsiri menggunakan metode destilasi uap air selama 4 jam. Uji aktivitas antibakteri menggunakan metode kertas cakram. Cawan petri berisi NA (masing-masing 10 mL) diinokulasikan strain bakteri 108 CFU/mL. Konsentrasi masing-masing minyak atsiri yaitu 5%; 2,5%; 1,25%; 0,625%; 0,313%; 0,156%. Hasil penelitian menunjukkan kadar minyak atsiri daun kayu manis sebesar 0,21% (v/b) dan rimpang temulawak sebesar 0,17% (v/b). Komponen minyak atsiri yang dianalisis dengan menggunakan GC-MS dengan konsentrasi tertinggi adalah senyawa alpha-pinene dan alpha curcumene. Hasil uji menunjukkan bahwa minyak atsiri daun kayu manis, rimpang temulawak, dan kombinasi memiliki diameter hambat minimum sebesar 10,53±1,03 mm (0,625%), 10,33±0,2 mm (0,625%), 9,40±0,30 mm (0,313%) terhadap bakteri Staphylococcus aureus dan 7,43±0,21 mm (0,625%), 9,17±0,28 mm (0,625%), 7,20±0,28 mm (0,625%) terhadap bakteri Escherichia coli. Hasil uji KHM kombinasi minyak atsiri daun kayu manis dan rimpang temulawak sebesar 0,313% pada bakteri Staphylococcus aureus dan 0,625% pada bakteri Escherichia coli.
