Abstract

Vibriosis merupakan suatu penyakit yang disebabkan oleh infeksi patogen golongan Vibrio yang dapat mengakibatkan kematian ikan mencapai lebih dari 80% pada budidaya ikan. Tujuan dari penelitian ini adalah menganalisis isolat patogen Vibrio spp yang menginfeksi ikan budidaya laut dengan beberapa set primer spesifik dan mengidentifikasi jenis bakteri Vibrio spp yang menginfeksi ikan laut budidaya di BBPBL Lampung. Koleksi isolat bakteri pada penelitian ini berjumlah 20 sampel isolat dari media selektif Vibrio TCBS dan 2 sampel isolat dari medium pengkaya TSB 2% NaCl. Identifikasi secara molekular menggunakan primer 16S rRNA untuk mengidentifikasi bakteri Vibrio sp panjang bandnya 700bp, primer Collagenase untuk mengidentifikasi bakteri V.alginolyticus panjang bandnya 737bp, primer Tdh untuk mengidentifikasi bakteri V. parahaemolyticus panjang bandnya 373bp, primer Tox R untuk mengidentifikasi bakteri V. parahaemolyticus panjang bandnya 368bp dan primer Vvh untuk mengidentifikasi bakteri V.vulnificus panjang bandnya 386bp. Hasil perhitungan prevalensi dari 22 isolat yang diidentifikasi secara molekular menggunakan primer 16S rRNA yaitu 81,82%, primer Collagenase yaitu 27,27%, primer Tdh yaitu 0%, primer Tox R yaitu 9,1% dan primer Vvh yaitu 9,1%. Kata kunci : Ikan laut, metode molekular, set primer , vibriosis. Vibriosis is a disease caused by infection with the Vibrio sp. pathogen which can cause fish mortality to reach more than 80% in fish culture. The purpose of this study is to examine and identify isolates of pathogenic Vibrio spp that infect marine fish culture with some specific primer sets in Main Center for Marine Aquaculture in Lampung. The collections of bacterial isolates in this study consisted of 20 isolates samples from Vibrio TCBS selective medium and 2 isolates samples from TSB 2% NaCl enrichment medium. Molecular identification used 16S rRNA primers to identify Vibrio sp bacteria with a band length of 700bp, Collagenase primers to identify V. alginolyticus at 737bp band length, Tdh primers to identify V. parahaemolyticus at 373bp band length, Tox R primers to identify V.parahaemolyticus has a band length of 368bp and primer Vvh to identify V.vulnificus with a band length of 386bp. The results of the total prevalence of 22 isolates, identified molecularly using 16S rRNA primers were 81.82%, Collagenase primers were 27.27%, Tdh primers were 0%, Tox R primers were 9.1% and Vvh primers were 9.1%.
