Abstract

DETERMINATION OF ANTIOXIDANT ACTIVITY IN THE ETHANOL EXTRACT OF JERUJU FRUIT (Acanthus ilicifolius L.) USING THE DPPH METHOD (1,1-diphenyl-2-picrylhydrazyl) Susan Brades 08061181621101 ABSTRACT Jeruju (Acanthus ilicifolius L.) is a mangrove plant that is widely used as an ingredient in traditional medicines to treat asthma, diabetes, hepatitis, as an anti-inflammatory and rheumatoid. Jeruju fruit is used as medicine for ulcers by the villagers of Sungai Lumpur, South Sumatera. As an antidote for snake bite poisoning in Kalimatan and cancer treatment by the people of West Sumatera. Jeruju plant has been known to contain flavonoid compounds as free-radical scavengers. This study was designed to evaluate and determined the phytochemical composition, extract characterization, total flavonoid content and antioxidant activity in the ethanol extract of Acanthus ilicifolius fruit. Phytochemical screening ensured the presence of steroids, triterpenoids, phenol, flavonoids, tannins and saponins. The determination of total flavonoids carried out by the colorimetric-AlCl3 method using a spectrophotometer UV-Vis while the antioxidant activity was determined using the DPPH method (1,1-diphenyl-2-picrylhydrazyl). The results of the total flavonoid content 9,329 ± 0,331 mg QE / g extract and antioxidant activity of IC50 217 ± 8,272 µg / mL can be classified as medium. MENGGUNAKAN METODE DPPH (1,1-diphenyl-2-picrylhydrazyl) Susan Brades 08061181621101 ABSTRAK Jeruju (Acanthus ilicifolius L.) merupakan salah satu tumbuhan mangrove yang banyak dimanfaatkan sebagai bahan obat tradisional untuk mengobati penyakit asma, diabetes, hepatitis, sebagai antiinflamasi dan pengobatan rheumatoid. Buah jeruju dimanfaatkan sebagai obat bisul oleh masyarakat Desa Sungai Lumpur, Sumatera Selatan. Sebagai penawar racun gigitan ular oleh masyarakat Kalimatan dan pengobatan kanker oleh masyarakat Sumatera Barat. Tumbuhan jeruju telah diketahui mengandung senyawa flavonoid sebagai penangkal radikal bebas. Penelitian ini bertujuan untuk mengetahui dan menentukan kandungan senyawa fitokimia, karakterisasi, kadar flavonoid total dan aktivitas antioksidan dari ekstrak etanol buah jeruju. Skrining fitokimia ekstrak etanol buah jeruju menunjukkan adanya kandungan steroid, triterpenoid, fenol, flavonoid, tannin dan saponin. Penentuan kadar flavonoid total dilakukan dengan metode kolorimetri-AlCl3 menggunakan spektrofotometer UV-Vis sedangkan aktivitas antioksidan ditentukan dengan metode DPPH (1,1-difenil-2-pikrilhidrazil). Hasil penentuan kadar flavonoid total sebesar 9,329 ± 0,331 mg QE/g ekstrak dan aktivitas antioksidan dengan nilai IC50 sebesar 217 ± 8,272 µg/mL termasuk dalam kategori antioksidan sedang.
