Abstract

The richness of Indonesian flora biodiversity makes Indonesian society utilize plants into drugs that are believed to treat disease hereditary in the local area. Diseases that are often found in people are generally degenerative diseases caused by oxidative stress. Oxidative stress is an imbalance of the number of free radicals and the number of antioxidants in the body. Plant The Asteraceae family is known for its benefits as anti-inflammatory, anticancer, antibacterial, and antioxidant. Kirinyuh leaves (Eupatorium inulifolium Kunth.) included to the Asteraceae family which has acted as an antioxidant and reported to have very strong antioxidant power, but not yet known to the classification of antioxidant compounds and isolation of antioxidant compounds. This study aims to determine the antioxidant activity of each fraction, classification of antioxidant compounds, and IC50 value of antioxidant compounds isolated from kirinyuh leaves. The research was conducted from October 2020 to March 2021, sampling is located in Bukit Sentiong, Koto Renah, Sungai Penuh City, Kerinci Regency, Jambi Province. Research method using of exploration experiment method is the research procedure begins with kirinyuh leaves sample preparation, then extraction, fractionation by method liquid-liquid fractionation, then purified and isolated with column chromatography and tested the antioxidant activity of the compounds by the method DPPH. The extraction results obtained the yield of kirinyuh leaves methanol extract as much as 24.61%. Fractionation results were obtained the extract yield of each fraction of n-hexane, ethyl acetate, and water-methanol respectively 33.66%, 41.66%, and 24.66%. The results of the antioxidant activity fraction kirinyuh leaves using thin-layer chromatography showed the fraction n-hexane and ethyl acetate fraction have relatively strong antioxidant activity compared to the methanol fraction of water. The result of purification and isolation geted pure isolates N1, N2, E1, E2, E6 and E8. isolates N1, N2, E1, and E2 is a group of terpenoid compounds that have different values of Rf, isolate E6 namely the flavonoid compound group, and E8 isolate, namely the tannin compound group. The antioxidant activity test using the DPPH method showed that isolates were N1 and E1 isolate had a very strong antioxidant activity with an IC50 value of 46.4925 ppm and 43.1465 ppm. Isolate N2, isolate E2, and isolate E6 have strong antioxidant activity with IC50 values of 88.4562 respectively ppm, 66.0675 ppm, and 62.4555 ppm. E8 isolate has an antioxidant activity which is being with an IC50 value of 107.5823 ppm. The conclusions obtained from this study were the groups of antioxidant compounds from the n-hexane fraction and the ethyl acetate fraction of kirinyuh leaves (Eupatorium inulifolium Kunth.) which were isolated, namely terpenoids, flavonoids and tannins which have very strong, strong and moderate antioxidant activity.
