Abstract

Free radicals cause damage to body cells, therefore antioxidants are necessary. Antioxidants are compounds that can prevent exposure to free radicals. Mahkota dewa (Phaleria macrocarpa (Scheff.) Boerl.) is one of Indonesia's native plants, so it contains compounds in the form of alkaloid compounds, flavonoid compounds, saponin compounds, tannin compounds, resin compounds, phenolic compounds and terpenoids. These compounds will reduce free radicals. This study aims to determine which fractions have antioxidant activity, to determine the class of compounds and the IC50 value of the antioxidant compounds of the leaves of mahkota dewa (Phaleria macrocarpa (Scheff.) Boerl.). This research was conducted in October 2020 to March 2021. The sampling location was in the Social Institution for the Elderly of our Expectations and the Road Across East of Km 29 in the Weights Village, Sub-District of North Indralaya. The research methods used included extraction, fractionation, and fraction antioxidant activity test using layer chromatography method. thin, purification of compounds using column chromatography, classification of compounds, and determination of IC50 values using the DPPH method. The results obtained from this study were the extract yielded a yield of 25.93% methanol extract. The results of the liquid-liquid fractionation obtained the yield value of the n-hexane fraction of 25.95%, 41.80% of ethyl acetate fraction, and 32.23% of methanol: water. The results of the antioxidant activity test using TLC plate with n-hexane and ethyl acetate fraction had a stronger antioxidant activity than the methanol: water fraction. The purification results using column chromatography of the n-hexane fraction were obtained 2 pure isolates, namely isolates N.1, N.2, while pure isolates E.1, and E2 were obtained from ethyl acetate fraction. The results of determining the class of compounds in each pure isolate, it is known that isolates N1, N2, and E1 belong to the terpenoid group compounds, while E2 is a compound of the Flavanoid group. The results of the antioxidant activity test for isolate N.1 had an IC50 value of 45.0530 ppm, the antioxidant activity was very strong, isolate N.2 had an IC50 value of 62.5388 ppm, the antioxidant activity was strong, isolate E.1 had an IC50 value of 190.6675 ppm antioxidant activity is classified as weak, isolate E.2 has an IC50 value of 77.8355 ppm, which is classified as strong antioxidant activity. The conclusion of this study is the fractions of n-hexane and ethyl acetate of the leaves of mahkota dewa (Phaleria macrocarpa (Scheff.) Boerl.) Contain terpenoid and flavonoid compounds and have antioxidant activity classified as very strong, strong and weak.
