Abstract

Gambir water extracts are known to contain catechins which have been proven as effective antioxidants. In order to increase the efficacy and also the permeability the penetration of catechins are turned into hydrophilic vesicles, such as transfersome dosage form. This dosage form can increase the penetration towards the stratum corneum. Transfersome preparation was formulated using factors of soy lecithin and polysorbate 80 as factor with 32 factorial designs in Design Expert®10 program. The effect of factor and compositions interaction were then observed based on the results of responses such as %EE, concentration after stability, PDI, as well as the pH. The observed test result were then analyzed using Design Expert®10 program in order to obtain the optimum formula. The optimum formula was concluded to be soy lecithin of 0.6 and polysorbate 80 of 0.07 g. This resulted into the desirability 0.589. The increase of soy lecithin resulted into a higher %EE and PDI as well as decrease of concentration after the stability test, PDI, and pH. After the formula was analyzed, optimum formula was obtained with the value of as such 98.826 ± 0.002%, 0.394, 212.8 nm, -28.2 mV, and 2.5 cP. After the stability assay of heating cooling using six cycles with different temperature of 4 and 40ºC, the result was more stable than the pure extracts. Diffusion analysis of the formula resulted into a higher diffuse percentage compared to the extract by itself which was 39.454 compared to 23.253%, this indicate that the transfersome has greater ability of penetration compared to the pure extract of gambir. Compartmental analysis using the WinSAAM® program followed the lag time model of 3 compartment with the correlation between the Qc and Qo value of greater than p-value of >0.05. Interaction analysis was conducted using the FTIR model using the placebo as well as the optimum formula, which result indicates that is no chemical interaction between the formula as well as the polymer compared to the placebo which showed that it is stable and it is applicable to be formulated into transfersome. TEM analysis result showed spheric form which will result in a higher diffuse percentage from the membrane. Based on these result, the water extract is suitable to be made in transfersome because it creates a stable spheric vesicle and which will resulted into a higher diffusion percentage compared just the pure form of extract.
