Abstract

Onion plant (Allium cepa L) is one of the traditional medicine from Amaryllidaceae family. The phytochemical screening showed that onion positive containing phenolic, flavonoid, and saponin compounds. The purpose of this research is to determine the anti bacterial activity of onion against S taphylococcus aureus and Escherichia coli b acteri a and to determine the total flavonoid content of the onion ethanolic extract. Simplicia was extracted by the maceration method with ethanol and then fractioned with ethyl acetate and ethanol water by liquid liquid partition method. After that the extracts were tested antibacterial activity by the paper disc diffusion method. The result showed that the diameter of the bacteriostat ic circle of the extract of ethanol, ethyl acetate fraction, and ethanol water fraction of onion was 4,52 mm, 5,3 mm , and 3,65 mm against S taphylococcus aureus bacteria but couldn’t inhibit the growth of Escherichia coli bacteria. The determination of the Minimum Inhibitory Concentration (MIC) was performed by liquid dilution method, while the Minimum Bacteri c idal Concentration (MBC) was performed by the solid dilution method. The result showed that the MIC value of the extract of ethanol, ethyl acetate fraction, and ethanol water fraction of onion was 6,25%, but the MBC value has not been obtained yet. The determination of the total flavonoid content of ethanolic extract was conducted using spectrophotometer based on the colorimetry method with AlCl 3 . The result showed that the total flavonoid content of the onion ethanolic extract wa s 53,4 mg Q E/g extra ct.
