Abstract

Bakteri Escherichia coli merupakan penyebab diare terbesar kedua setelah rotavirus di dunia. E. coli ditemukan dalam saluran pencernaan sebagai flora normal manusia dan hewan. E. coli menjadi patogen jika jumlah bakteri dalam saluran pencernaan meningkat. Daun beluntas mengandung senyawa antibakteri sekaligus antidiare yaitu tanin. Penelitian menggunakan tikus putih jantan galur wistar yang diinduksi bakteri E. coli. Kelompok perlakuan dibagi menjadi enam: kelompok normal, kelompok kontrol negatif, kelompok kontrol positif, dan tiga kelompok uji (dosis 125, 250, dan 500 mg/kgBB). Tikus yang terinfeksi E. coli diberi perlakua: kontrol negatif diberikan Na-CMC, kontrol positif diberikan gentamisin, dan uji diberikan suspensi ekstrak sesuai dosis. Uji aktivitas antidiare dilakukan tiga hari dengan pengamatan: waktu awal terjadi diare, konsistensi feses, frekuensi diare, perubahan berat badan, efek antidiare, dan jumlah koloni E. coli pada feses. Kadar tanin total yang didapat sebesar 16,4%. Hasil penelitian in vivo menunjukan dosis 125, 250, dan 500 mg/kgBB memiliki persen efek antidiare berturut-turut 36,35%, 42,69%, dan 50,19%. Jumlah koloni E. coli saat penyembuhan diare dosis 125, 250, dan 500 mg/kgBB berturut-turut 112,8 x 105 (cfu/g); 109,2 x 105 (cfu/g); dan 104,6 x 105 (cfu/g). Ekstrak etanol dosis 500 mg/kgBB mengurangi populasi E. coli yang hampir sama dengan kontrol positif, gentamisin (P>0,05).
