Abstract

VCO is a functional food that is often consumed as an alternative in improving health. In the body there are probiotic bacteria which are normal microflora of the intestine, an example of probiotic bacteria is L. bulgaricus. This research was conducted to see the effect of VCO on the activity of L. bulgaricus probiotic bacteria in the body. The growth (viability) test of L. bulgaricus was carried out by counting the colonies used a total plate count method. Diffusion method was used to determine the antibacterial activity of L. bulgaricus metabolites. MIC determination used the dilution method (microdilution). VCO was made using the fishing method and has met the standards for specific gravity (0.9149), refractive index (1.4546) and water content (0%). The fatty acid component was tested using GCMS with the highest concentration, namely lauric acid (17.92%). The VCO concentration used in the bacterial growth (viability) test was 1%, 5%, 10%, 15%. The best concentration of bacteria was at concentration of 1% with p<0,05. There was a significant difference in the number of probiotic bacteria in the concentration of 15% with negative and positive controls. The number of bacteria at the four concentrations were 150.8 x 1012, 111.4 x 1012, 109.25 x 1012, 55.667 x 1012, respectively. Antibacterial activity tested of L. bulgaricus metabolites with three concentrations (100%, 75%, 25 %) resulted in the resistance response obtained, namely weak-moderate. In the antibacterial activity test on L. bulgaricus metabolites that were not treated with VCO produced a significant difference where the p<0,05. Testing the Minimum Inhibitory Concentration (MIC) on L. bulgaricus metabolites that had been treated with VCO and not treated resulted in different MIC. The MIC on untreated metabolites was 4.69%, while the MIC on treated metabolites was 9.38%. ABSTRAK VCO merupakan pangan fungsional yang sering dikonsumsi untuk dijadikan sebagai alternatif dalam meningkatkan kesehatan. Didalam tubuh terdapat bakteri probiotik yang merupakan mikroflora normal usus, contoh bakteri probiotik itu ialah L. bulgaricus. Penelitian ini dilakukan untuk melihat pengaruh VCO terhadap aktivitas bakteri probiotik L. bulgaricus yang ada didalam tubuh. Uji pertumbuhan (viabilitas) bakteri L. bulgaricus dilakukan dengan menghitung koloni menggunakan total plate count. Penentuan aktivitas antibakteri metabolit L. bulgaricus menggunakan metode difusi. Penentuan KHM menggunakan metode dilusi (microdilution). VCO dibuat menggunakan metode pancingan dan telah memenuhi standar bobot jenis (0,9149), indeks bias (1,4546) dan kadar air (0%). Komponen asam lemaknya diuji menggunakan GCMS dengan konsentrasi tertinggi yaitu asam laurat (17,92%). Konsentrasi VCO yang digunakan pada uji pertumbuhan bakteri (viabilitas) yaitu 1%, 5%, 10%, 15%. Konsentrasi yang paling bagus jumlah bakterinya berada pada konsentrasi 1%. Terdapat perbedaan yang signifikan pada jumlah bakteri probiotik konsentrasi 15% dengan kontrol negatif maupun positif ditandai dengan nilai p<0,05. Jumlah bakteri pada keempat konsentrasi itu berturut-turut yaitu 150,8 x 1012, 111,4 x 1012, 109,25 x 1012, 55,667 x 1012. Pengujian aktivitas antibakteri metabolit L. bulgaricus dengan tiga konsentrasi (100%, 75%, 25%) menghasilkan respon hambatan yang didapat yaitu lemah-sedang. Pada uji aktivitas antibakteri pada metabolit L. bulgaricus yang tidak diberi perlakuan dengan VCO menghasilkan perbedaan yang signifikan dimana nilai p<0,05. Pengujian Konsentrasi Hambat Minimum (KHM) pada metabolit L. bulgaricus yang telah diberi perlakuan dengan VCO dan tidak diberi perlakuan menghasilkan KHM yang berbeda. KHM pada metabolit yang tidak diberi perlakuan yaitu 4,69% sedangkan KHM pada metabolit yang diberi perlakuan yaitu 9,38%.
