Abstract

Based on the color pigment, rice has a variety of colors. Red rice is a colored rice with a higher genetic variation than other colored rice. Genetic conservation is an effort to prevent the loss of local red rice genetic resources by providing superior varieties that have more profitable characters for farmers. One of the genetic conservation efforts that must be done is DNA sequencing using a molecular approach. DNA barcoding is one of the molecular approaches used to speed up and simplify the process of identifying species before extinction occurs. Source of DNA Barcoding sequences in plants can be obtained from chloroplast DNA (cpDNA). The matK gene is a sequence in chloroplast DNA that can be amplified with a high success rate, has a high substitution rate, has a better distinguishing ability in research on intraspecies variation and is the most varied gene in angiosperms so it is appropriate if it is used as a barcode. This study aims to Barcoding the DNA of local varieties of red rice in South Sumatra based on the matK gene. This research was conducted from January to April 2021 at the Laboratory of Genetics and Biotechnology, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University. The methods used in this study included DNA isolation using Tiagen Plant Genomic DNA KIT DP305, isolated electrophoresis, DNA amplification using matk Xf and matK-MALP primers and DNA sequencing. The result showed that the matK gene in 8 samples of local varieties of South Sumatra red rice was successfully amplified by the presence of DNA bands at size between 750 bp – 1000 bp and there was a high similarity with Oryza sativa maturase K gene No. Access KU923990.1 but there are differences in nucleotide bases between sample local varieties of South Sumatra red rice as indicated by mutation events.
