Abstract

Background: One of the factors that have the potential to cause root canal treatment failure (RCT) is the remaining bacteria in the root canal. Efforts to prevent RCT failure are carried out by using irrigation solutions with antibacterial properties. Chlorhexidine is an irrigation solution that has antibacterial properties that are most commonly used, but its cytotoxicity is still high. Curcuma zedoaria contains antibacterial substances that have the potential as an alternative irrigation solution. The purpose of this study was to determine the antibacterial effect of extract C. zedoaria in the root canals of Wistar rats. Methods: This study used a laboratory experimental study with in vivo post test only control group design. C. zedoaria extract was prepared and diluted to concentrations of 50%, 25%, and 12.5%. RCT was performed on 25 adapted rats. The irrigation solution used was C. zedoaria extract with various concentrations, CHX 2%, and aquadest. The antibacterial test was carried out by counting the bacterial colonies formed on the agar media using the Colony Count application. Data analysis was carried out with the One Way ANOVA test. Result: The results of the antibacterial test showed the lowest average number of bacterial colonies was in the C. zedoaria 12.5% extract treatment group ​​and the highest was in the 50% C. zedoaria treatment group. One Way ANOVA test results obtained a significance value of 0.410 (p>0.05), indicating that there was no difference in the number of bacterial colonies between each research group. Conclusion: The ​​ extract of C. zedoaria 12.5% had a higher antibacterial effect than the positive control. The extract of C. zedoaria 50% and 25% had lower antibacterial effect than the negative control. There is no significant difference in antibacterial effectiveness between each treatment group.
