Abstract

Daun jeruju (Acanthus Ilifolious L.) merupakan salah satu tumbuhan mangrove sejati yang digunakan sebagai bahan obat dalam mengobati penyakit diare. Penelitian ini bertujuan untuk melihat pengaruh ekstrak daun jeruju terhadap pertumbuhan dan aktivitas antibakteri probiotik L. bulgaricus. Hasil uji karakterisasi ekstrak daun jeruju didapatkan kadar air (6 ± 1), bobot jenis ( 0,6 ± 0,005), susut pengeringan (6,38 ± 1,13) dan kadar abu total ( 2 ± 0,5). Penentuan kadar flavonoid total sebesar 21,003 mgQE. Pengujian uji pertumbuhan bakteri probiotik dengan penambahan ekstrak daun jeruju digunakan metode total plate count. Hasil yang didapat pada uji pertumbuhan terbaik pada konsentrasi 15% dengan jumlah koloni 4,95 x 1012 cfu/mL. Uji aktivitas antibakteri menggunakan metode difusi cakram kertas dengan variasi konsentrasi 25%, 75%, 100% kontrol positif antibiotik Ciprofloxacin dan kontrol negatif pelarut aquadest. Hasil uji aktivitas antibakteri metabolit L. bulgaricus menunjukkan bahwa kelompok uji pertumbuhan dan kelompok kontrol positif uji pertumbuhan pada konsentrasi 100% sudah menunjukkan respon hambatan pada bakteri Escheria coli dengan kriteria sedang.
