Abstract

Introduction: Diabetes can be treat with a plant that has a potential to be anti diabetes which is the (Mutingia calabura L.). The leaves of the Muntingia is contain steroid and flavonoid. Flavonoid helps to control alpha-glucosidase enzymes whereas steroids can increase the blood sugar level. A previous studied done by Putriana, 2019 that the fraction of Muntingia leaves did not actively inhibit the alpha-glucosidase enzyme which is 106,333. Therefore, further research about the potential nonpolar fractions extract of Ethyl acetate from the Muntingia leaf in inhibiting the alpha-glucosidase enzyme is required. Methods: Ethyl acetate extract by simple maceration is continued with the fractionation process using a mixture of n-hexane solvent and ethyl acetate with 5 different concentrations. This studied about the inhibition of alpha-glucosidase enzyme was done on all fractions and control positive acarbose by measuring the absorbable rate of p-nitrofenil-alpha-D-glukopiranosa using spectrophotometer with length of 400nm. The absorbable rate used to count the percentage of alpha-glucosidase enzyme inhibition. Linier regression analysis was be done on the results of the enzyme inhibition to get a value of IC which then be categorized into very active, active and non active inhibition. Results: Nonpolar fraction extract of ethyl acetate from Muntingia leaves contain active steroid and flavonoid compounds. Effect of active inhibition by nonpolar fraction extract of ethyl acetate which worked as anti diabetes medication is the n-hexane solvent: ethyl acetate F5 (1:9). This due the higher ethyl acetate solvent compared to n-hexane and also a higher constant rate compared to the other four fractions. The effect of non active inhibition by nonpolar fraction extract of ethyl acetate from Muntingia leaves is F1 (9:1), F2 (7:3), F3 (5:5) dan F4 (3:7). Conclusion: The level of secondary metabolite found in nonpolar fraction extract of ethyl acetate from Muntingia leaves is steroid and flavonoid. Nonpolar fraction extract of ethyl acetate is unable to hinder the alpha-glucosidase enzyme. Keywords: Muntingia leaves, Dendrophthoe pentandra (L.)
