Abstract

The position of Pulau Payung, which is right in the middle of the Musi River estuary, makes this island potentially polluted by heavy metal waste. One alternative that can be used for the management of water pollution from heavy metals is phytoremediation. Avicennia alba has the ability to absorb heavy metals so that it can be used as a phytoremediation agent. The physiological response of Avicennia alba to heavy metal stress causes changes in the activity of free radical enzymes, especially PO and PPO, so a study was conducted to determine the activity of peroxidase (PO) and polyphenol oxidase (PPO) as a response to plant physiology adaptation of Avicennia alba in phytoremediation of heavy metals in Pulau Payung South Sumatra. This research was carried out in April 2021-July 2021 at the Physiology and Developmental Laboratory of the Department of Biology, Sriwijaya University. The sampling process uses the Cluster sampling method. Measurement of the levels of Pb and Cu using atomic absorption spectrophotometry. Protein extraction using the principle of centrifugation. Measurement of total protein content, and activity of PO and PPO using uv-vis spectrophotometry method. Analysis of the data used in the form of quantitative data for measuring pH, temperature, metal content of Pb and Cu and the activity data of PO and PPO presented by analysis of data centering and standard deviation. Based on the research that has been done, it is found that the high activity of PO and PPO in A. alba in phytoremediation of heavy metals in Payung Pulau at station 2, namely PO of 59.64 ± 10.60 U/mg protein/minute and PPO of 117.94 ± 6.21 U/mg protein/minute indicates a physiological response that is influenced by the high content of heavy metals Cu and Pb in the sediment with an average of Cu 6.71 ± 0.08 mg/kg and Pb of 6.98 ± 0.57 mg/kg.
