Abstract

Latar belakang: Senyawa metabolit sekunder yang diduga dapat bekerja sebagai antihiperurisemia adalah saponin, tanin, flavonoid, terpenoid, serta alkaloid protoberberin seperti berberin, jatrorizin, dan palmatin. Diketahui bahwa tanaman akar kayu kuning (Arcangelisia flava (L.) Merr.) memiliki kandungan metabolit sekunder seperti saponin, tanin, terpenoid, flavonoid, dan alkaloid, sehingga memiliki potensi antihiperurisemia. Tujuan: Untuk mengetahui kemampuan ekstrak etanol akar kayu kuning (Arcangelisia flava L. Merr) dalam menghambat enzim xantin oksidase. Metode: Jenis penelitian ini adalah eksperimental laboratorium secara in vitro dengan menggunakan rancangan post-test. Proses ekstraksi akar kayu kuning dengan metode maserasi. Uji inhibisi xantin oksidase dilakukan dengan menggunakan spektrofotometri UV-Vis dengan 4 konsentrasi ekstrak etanol akar kayu kuning dan allopurinol digunakan sebagai pembanding. Hasil: Ekstrak etanol akar kayu kuning mengandung senyawa alkaloid, flavonoid, triterpenoid, dan kuinon. Ekstrak etanol akar kayu kuning mempunyai IC50 sebesar 30,4365 ppm dan tergolong aktif dalam menghambat enzim xantin oksidase. Kesimpulan: Ekstrak etanol akar kayu kuning mempunyai potensi sebagai antihiperurisemia melalui inhibisi enzim xantin oksidase. Kata kunci: Kayu kuning (Arcangelisia flava L. Merr), potensi antihiperurisemia, inhibisi enzim xantin oksidase, fitokimia.
