Abstract

The COVID-19 pandemic is still a global problem, therefore vaccination programs need to be continuously improved and developed to overcome it. Research of the immune system genes can help predict the outcome condition of a disease. IFNL4 (regulated by rs368234815), as the foremost protector of the respiratory epithelial barrier, has great potential in its role in treating this disease. Information on local characteristics is hoped to improve health programs in the area. This study aims to determine the rs368234815 genotype description of the people of Palembang city and also their antibody levels after the second SARS-CoV-2 vaccination. This research is an observational descriptive study with primary data. The sample from this study is the people of Palembang city who had vaccinated the second dose of SARS-CoV-2 (Bio Farma/Sinovac, inactivated) in August 2021. Polymorphism data was obtained by PCR-RFLP, and antibody levels by measuring IgG s1 RBD SARS-CoV- 2. The data is used to see the frequency distribution of the rs368234815 polymorphism genotype and its antibody levels. Of the 86 samples, 94.19% had the TT/TT genotype, only 5.81% had the TT/ΔG genotype, and ΔG/ΔG was not found. The TT/TT group had a higher mean antibody level than the TT/ΔG group (1,686.89 AU/ml: 858.96 AU/ml), and in all groups, more low-category antibody levels were found (<840 AU/ml). The majority rs368234815 genotype of the people of Palembang city is TT/TT, with the majority of antibody levels after the second SARS-CoV-2 vaccination beign relatively low.
