Abstract
The endophytic microorganism is a form of bacteria or a fungi microorganism that form colonies interiororgans of plants, but does not harm the host plant. The secondary metabolites compound heve been isolated from endophytic fungi (Penicillium citrinum) living in symbiotic dill for malaria that is brotowali ( Tinospora crispa L ). Isolation begin with cultivation of Penicillium citrinum fungi in 2 liter of PDB's media ( Polato Dextrose Broth ) for four weeks. Media is extracted into the solvent n-heksan and ethyl acetate following by evaporation. Ethyl acetate extracts were separated by chromatography techniques in order to get pure compound in the form of yellow crystal with melting point 197-198 °C. Phytochemical tests showed that the isolated compound is the alkaloid. The UV spectrum of the isolated compound showed maximum absorption at (log a): 315nm (4,38), 253nm (4,79), dan 213 nm (5,33). The 'H-NMR spectrum showed that seven proton signal which is three signals for methyl proton at Sn 1,33; 1,21 and 2,01 ppm, two signals for methine sp3 proton at SH 2,97 and 4,77 ppm, one signal for methine sp2 proton at SH 8,23 ppm, and one proton signal is very low field which at SH 15,10 ppm. The 13C-NMR spectrum showed that the result of isolation compound has 13 carbon atoms. The identification signals of carbon and proton further defmed based on 2D NMR spectra. Base on of all the spectroscopy correlation data UV, 'H-NMR, 13CNMR, HMQC, HMBC, and COSY showed that result of isolation compound is alkaloid tetrahydroisoquinolin’s type, that is acid-7-hydroksi-3,4,5-trimethyl-6-on-2,3,4,6-tetrahydroisokuinolin-8-oat with molecule formula C13H15O4N (Mr= 249). The result of isolation compound showed that activity as anti malarial to Plasmodium falciparum 3D7, and assigns value 1C50 0,025 pg/L or 0,10 pM.
