Abstract

Background: Leprosy is a chronic infectious disease caused by Mycobacterium leprae. Transmission of leprosy occurs through direct contact with sufferers. Leprosy is an immunologic disease, where the pathogenesis of the disease is determined by the condition of the body's immune system. IL-10 has a pathological impact on leprosy. Polymorphisms in the IL-10 gene promoter affect the amount of IL-10 secretion in the body. The amount of IL-10 secretion will affect the susceptibility and severity of leprosy in an individual. The purpose of this study was to identify the -2763 point polymorphism of the IL-10 gene promoter in leprosy patients at dr. Mohammad Hoesin Palembang. Methods: This research is a descriptive observational study with a cross-sectional design. Identification of polymorphisms was carried out on DNA extracts of leprosy patients using the PCR-RFLP method. This study only observed and presented the results of the observation of the -2763 point polymorphism of the IL-10 gene promoter. Results: Of the 48 samples of leprosy patients identified, the majority were aged <50 years (72.91%), male (66.7%), with multibacillary leprosy (91.7%), and the majority belonged to the ethnic group. Malay-South Sumatra (52.2%). From 48 samples of leprosy patients, the frequency of the genotype CC was 60.4%, AC was 39.6%, and there was no genotype AA. The frequency of the C allele was found as much as 80.2% and the A allele as much as 19.8%. Conclusion: The majority of leprosy patients at dr. Mohammad Hoesin Hospital for the period January–February 2019 had wild-type (CC) genotypes. Keywords: Leprosy, polymorphism, IL-10, point -2763
